Kjeldahl technique, is a worldwide accepted
technique for more than 100 years for the estimation of nitrogen and protein.
Digested samples are distilled with sodium hydroxide, which converts ammonium
sulphate to ammonia. The amount of ammonia present and the amount of nitrogen
present in the sample is determined by titration. Distillation systems follows the above
principle of operation with little modernization. Instead of traditional
heating of kjeldahl flasks, the borosilicate glass tubes with digested sample
are purged with steam after automatic addition of sodium hydroxide. The
liberated ammonia is captured in boric acid that is kept in a conical flask for
further titration.

Analyzer is recommended to be used to measure the nitrogen content in a protein
containing sample, from this measured nitrogen, protein content in the sample
can also be estimated.
TKN measurement mainly involves three steps.,
viz., digestion of the sample, distillation of digested sample and lastly
titration. Except digestion, distillation and titration steps are automated in
this analyser.
